Посттравматический остеоартроз (ПТОА) суставов является многофакторным заболеванием, обусловленным механическими и биологическими изменениями; оно, в основном, характеризуется дегенерацией суставного хряща, субхондральной кости и изменением свойств синовиальной жидкости, вязкость которой снижается, что требует коррекции. Биоактивные вещества, состоящие из полинуклеотидов, попадая в сустав, расщепляются до простых нуклеотидов, нуклеозидов и азотистых оснований, которые, как известно из литературы, в физиологических концентрациях присутствуют во внеклеточной среде, и представляют собой фундаментальные субстраты для регенерации клеток сустава.</p><p>Цель исследования – оценить влияние полинуклеотидов в качестве инъецируемых пролиферантов на проявления экспериментального ПТОА.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Работа была выполнена на белых нелинейных крысах самцах. Интенсивность перекисного окисления липидов оценивали по уровню накопления в сыворотке крови животных гидроперекисей липидов (ГПЛ), об активности антиоксидантной системы судили по тиоловому статусу (ТС). Для оценки синтетической активности остеобластов определяли содержание остеокальцина (ОК), а для метаболизма суставного хряща – гиалуронана (ГН).</p></sec><sec><title>Результаты</title><p>Результаты. На 7-е сутки экспериментальной работы нами было подтверждено формирование модели ПТОА на основании изменений уровней маркеров сыворотки крови. В результате внутрисуставных инъекций пролиферантов на основе полинуклеотидов на 28-е сутки эксперимента отмечали повышение ГПЛ, в то время как ТС был снижен. Под действием полинуклеотидов дисбаланса в механизмах регуляции костно-хрящевого компонента сустава не наблюдалось, что подтверждено снижением показателей ОК и ГН на 28-е сутки эксперимента против группы сравнения.</p></sec><sec><title>Заключение</title><p>Заключение. Данные проведенного эксперимента свидетельствовали о начале дегенеративных изменений на 7-е сутки после моделирования ПТОА. Полинуклеотиды, инъецируемые в качестве пролиферантов, представляют собой вещества способные поддерживать процессы физиологического восстановления костно-хрящевых структур и окислительно-восстановительной реакции суставных тканей при экспериментальном ПТОА.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Relevance</title><p>Relevance. Post-traumatic osteoarthrosis (PTOA) is a multifactorial disease caused by mechanical and biological changes. It mainly features the degeneration of the articular cartilage and subchondral bone, and changes in the properties of synovial fluid; its viscosity decreases which requires correction. When entering the joint, the bioactive substances consisting of polynucleotides break into simple nucleotides, nucleosides, and nucleobases, which, as is known from the published studies, are present in the extracellular environment in physiologic concentrations, and are fundamental substrates for joint cell regeneration.</p><p>The objective of this study is to investigate the impact of polynucleotides as injectable proliferants on the presentations of simulated PTOA.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The experiment involved white outbred male rats. The intensity of lipid peroxidation was assessed by lipid hydroperoxides (LHP) accumulation in the serum of the animals, and antioxidant system activity by thiol status (TS). To assess the synthetic osteoblasts activity osteocalcin (OC) content was determined, and the articular cartilage metabolism was defined by hyaluronan (HN) content.</p></sec><sec><title>Results</title><p>Results. On Day 7 of the experiment, the PTOA simulation was confirmed by the changes in the levels of serum markers. Resulting from intra-articular administration of polynucleotide-based proliferates on Day 28 the decrease in HPL was observed while TS was increased. The polynucleotides didn’t cause any imbalance the regulation mechanisms of the joint osteochondral component which was confirmed by a decrease in OC and HN values on Day 28 versus the comparison group.</p></sec><sec><title>Conclusion</title><p>Conclusion. The experiment findings suggest the onset of degenerative changes as early as on Day 7 of PTOA simulation.
